慢性束缚应激对棕色田鼠和ICR小鼠情绪行为和学习记忆的影响

摘要:
运用旷场实验、水迷宫定位航行和空间搜索实验,观察慢性束缚应激对棕色田鼠行为表现和学习记忆能力的影响,并与ICR小鼠进行比较.结果显示:慢性束缚应激使棕色田鼠和ICR小鼠的体重增长率降低,旷场实验中的水平穿格数、垂直站立次数和修饰次数均明显减少(P<001).注射雌激素(苯甲酸雌二醇)可改善束缚应激引起的体重与行为变化,且ICR小鼠对雌激素的反应比棕色田鼠更为明显.束缚应激使棕色田鼠水迷宫实验中的潜伏期显著延长,原平台穿越次数和原平台象限时间比率显著降低(P<001).但应激使ICR小鼠潜伏期缩短,原平台穿越次数和原平台象限时间比率明显提高(P<001).注射雌激素可完全改善束缚应激引起的学习记忆能力的变化.结果表明,慢性束缚应激能引起棕色田鼠和ICR小鼠抑郁样行为表现.但对学习记忆的影响则相反,慢性束缚应激能降低棕色田鼠空间学习记忆能力,对ICR小鼠的空间学习记忆能力却有促进作用,雌激素能很好地改善应激对ICR小鼠行为的影响和对棕色田鼠空间学习记忆能力的影响.

Effects of chronic restraint stress on behaviors and learning-memory in female Microtus mandarinus and ICR mice

Abstract:
The open field was used to observe behavioral changes, and the place navigation and the spatial probe in Morris water maze were performed to determine learning-memory abilities.The results showed that,compared with the control group,CRS on both female Microtus mandarinus and ICR mice showed a significant reduction in body weights and decreased the locomotion,rearing and grooming in open field test (P<001). Compared with the CRS group, hyodermic injection of β-estradiol 3-benzoate(EB) into the animal receiving CRS-treated significantly increased body weight(P<005)and insignificantly increased behavioral ability, which included locomotion,rearing and grooming in open field test. Compared with the control, CRS on ICR-mice distinctly shortened the latency in place navigation test, while hyodermic injection of EB showed insignificant changes. As to the Microtus mandarinus in Morris water maze, compared with the control, CRS prolonged the latency markedly, but the combination of the injection of EB and the CRS obviously cut down the latency, and improved the times of crossing the exact position of the former platform and the time ratio of remaining in the former platform when compared with the control group and the CRS group. These results suggest that the learning-memory ability of Microtus mandarinus is reduced with CRS, but promotion on ICR mice. Estrogen can improve the stress on good behavior in ICR mice and the impact on learning and memory in Microtus mandarinus.
