丹参铜锌超氧化物歧化酶(Cu/Zn-SOD)基因的克隆与生物信息学分析

摘要:
依据已构建的丹参(Salvia miltiorrhiza Bunge) cDNA文库和降落PCR的方法获得丹参胞质铜锌超氧化物歧化酶(Cu/Zn-SOD)基因,并用生物信息学方法对该基因编码产物从氨基酸组成、理化性质、亲水性/疏水性、二级结构以及功能等方面进行了预测和分析.结果表明,克隆得到的丹参Cu/Zn-SOD位于细胞质中,是非跨膜的非分泌性亲水蛋白,主要由无规则卷曲和延伸链等蛋白质二级结构元件构成,与其他植物的胞质Cu/Zn-SOD在序列组成、结构及活性位点等方面均有高度的相似性.

Abstract:
Based on the constructed cDNA library, a Cu/Zn-superoxide dismutase gene (Cu/Zn-SOD) from Salvia miltiorrhiza Bunge was cloned by TD PCR. The characteristics including the composition of amino acid sequences, hydrophobicity or hydrophilicity, secondary structure of protein and function of the Cu/Zn-SOD were analyzed and predicted by the tools of bioinformatics.The results showed that SmCu/Zn-SOD is a hydrophilic, non-transmembrane and non-secretory protein. The main motif of Cu/Zn-SOD in secondary structure was random coils and extended strands.Despite sequence variations, the core structure, sequence and activity sites of SmCu/Zn-SOD were highly similar to that of other green plants.
