Cloning and bioinformatics analysis of Cu/Zn-SOD gene from Salvia miltiorrhiza Bunge

Abstract:
Based on the constructed cDNA library, a Cu/Zn-superoxide dismutase gene (Cu/Zn-SOD) from Salvia miltiorrhiza Bunge was cloned by TD PCR. The characteristics including the composition of amino acid sequences, hydrophobicity or hydrophilicity, secondary structure of protein and function of the Cu/Zn-SOD were analyzed and predicted by the tools of bioinformatics.The results showed that SmCu/Zn-SOD is a hydrophilic, non-transmembrane and non-secretory protein. The main motif of Cu/Zn-SOD in secondary structure was random coils and extended strands.Despite sequence variations, the core structure, sequence and activity sites of SmCu/Zn-SOD were highly similar to that of other green plants.
