丹参COR413基因克隆及表达分析

摘要:
从丹参EST数据库筛选出一条与冷调节基因(cold-regulated gene,COR)家族同源性较高的基因序列,并利用PCR方法得到该基因的DNA序列,命名为SmCOR413.该基因DNA序列长1708 bp,包含4个外显子和3个内含子,编码171个氨基酸,与其他9种植物中的COR413高度相似,相似性介于66%~78%之间,推测为COR413基因家族的一个新基因.生物信息学分析表明,SmCOR413所编码蛋白的分子量为19.77 kD,理论等电点为8.95,不具备信号肽.实时定量PCR检测的结果显示,SmCOR413在丹参根、茎和叶中都有表达,在叶、茎中表达量较高,根部的表达量最低.

Cloning and expression of SmCOR413 from Salvia miltiorrhiza Bunge

Abstract:
One sequence which showed homology with COR413(cold-regulated gene)gene family was found from the expressed sequence tags(EST) of Salvia miltiorrhiza using the BLAST approach. The full-length cDNA and DNA sequence of the gene was cloned by PCR and named as SmCOR413. The DNA sequence was 1708bp, and contained four extrons and three introns. It encoded a predicated polypeptide of 171 amino acids with a molecular weight of 19.77 kDa and a predicated pI of 8.95. Amino acids similarity of SmCOR413 compared with COR413 in other nine plants reached 66%—78%. This gene maybe belonged to COR413 gene family without signal peptide. Real-time PCR result showed that the SmCOR413 expressed in root, stem and leaf. The mRNA of SmCOR413 was the most abundant in leaf and stem, the least in root.
