A modified method of DNA extraction from Schisandra sphenanthera Rehd.et Wils for AFLP analysis

Abstract:
To obtain high quality genomic DNA isolated from dry leaves of Schisandra sphenantheraRehd et Wils for AFLP analysis. A modified CTAB method was developed, which includes dissolving plant tissue in CTAB-free buffer before isolating the nucleolus and removing polysaccharide with high salt. The results showed that the value of OD260/OD280 was from 1.80 to 2.00, main bands were clear, and degradation was nearly free. Genome DNA was suitable for digestion by restriction endonucleases (EcoR I and Mse I). AFLP analysis indicated the patterns were clear and polymorphic, indicating that this method could be used for extracting ideal DNA from dry leaves of Schisandra sphenanthera Rehd. et Wils and suitability for AFLP analysis.
