反相胶束体系中氯过氧化物酶催化氧化吲哚的“超活性”研究

摘要:
氯过氧化物酶(CPO)在阳离子表面活性剂(DTAB/CTAB)-异辛烷-水反胶束体系中,表现出“超活性”,其氧化活性可分别提高177%和156%.研究了表面活性剂浓度、水含量对CPO氧化活性的影响,并通过反相胶束与水相中的CPO与底物H2O2形成的化合物Ⅰ的Soret吸收光谱的对比,对其形成的中间体的稳定性进行了研究.结果表明,CPO的氧化活性对表面活性剂浓度及水含量均呈现“钟罩”型依赖规律,反相胶束体系中间体(化合物Ⅰ)的稳定性增强.

Study on the superactivity of oxidation of indole catalyzed by chloroperoxidase in reverse micelles

Abstract:
Chloroperoxidase (CPO) was found to have a superactivity in reverse micelles formed by DTAB /CTAB-isooctane-water . The oxidation activity of CPO was enhanced up to 177% and 156% in the DTAB /CTAB-isooctane-water media respectively. The effects of water content and surfactants concentration on the catalytic performance of CPO were investigated. Moreover, the intermediate stability was studied based on the comparison of the Soret band of CPO-H2O2 complexes in reversed micelles to that in aqueous system.It was found a “bell shaped”dependences between the surfactants concentration or water content and the oxidation activity CPO,and enhancement in the intermediate stability in reversed micelles solutions.
