苯酚-硫酸法测定太白泡沙参总多糖的含量

摘要:
采用水提醇沉法制备太白泡沙参总多糖储备液.在优化条件(包括检测波长、试剂用量、反应温度)下,建立了苯酚-硫酸体系测定太白泡沙参总多糖含量的方法,并对该方法的精密度、重复性和加样回收率进行了考察.结果表明,对照液葡萄糖浓度为0.1 mg/mL时,1.0 mL的对照液所需5 %苯酚的用量为1.0 mL,浓硫酸的用量为5.0 mL,反应温度为室温,在波长为490 nm处,回归方程Y=9.695X-0.009 2(r=0.999 8),葡萄糖的质量浓度在0.01~0.09 mg/mL线性关系良好.该方法的精密度RSD=0.66%(n=5),重复性RSD=0.29%(n=5),平均回收率为99.9%(n=9),RSD=2.1%.该方法简便、合理,适合太白泡沙参总多糖含量的测定.

Determination of the total polysaccharide in the A.potaninii Korsh. from Taibai Mountain in China with phenol-sulfuric acid method

Abstract:
The total polysaccharides in the root of the A.potaninii Korsh from Taibai Mountain in China was determinated. The stock solution of the total polysaccharides was prepared from the extraction with water, and then the extraction solution was treated by alcohol to eliminate protein. Based on the optimization of conditions including testing wave length, amoune of reagents and reaction temperature et al, the method of phenol-sulfuric acid was established for determination of the total polysaccharide in the A.potaninii Korsh. The tests induding accuracy, reproducibility and recovery rate have been carried out. When the concentration of control solution containing glucose was 0.1 mg/mL, the 1.0 mL of 5 %-phenol aqueous solution and 5.0 mL of concentrated sulfuric acid were necessary to complete the reaction at room temperature. The results are as follows. The RSD of accuracy was 0.66 % (n=5), RSD of reproducibility was 0.29 % (n=5), and average recovery rate was 99.9 % (n=9). The results indicated that the method is simple, reliable to determine the total polysaccharide in the A.potaninii Korsh.
