蚕蛹蛋白提取工艺优化的研究

摘要:
以蚕蛹为原料,选用5种不同溶剂作为提取剂,采用索氏提取法提取蛹油制备脱脂蛹;对脱脂蛹采用二次回归正交旋转组合设计,以碱溶酸沉法提取蚕蛹蛋白.结果表明,正己烷为蚕蛹油最佳提取溶剂,其出油率为28.9%,提取蚕蛹蛋白的最佳条件为pH=10,料液比1∶12,温度60℃,提取时间90 min,连续提取两次,提取率为48.22%;蚕蛹蛋白等电点为4.4,冻干后蛋白含量达到95.2%.

Study on the optimization of extraction technology of silkwormpupa protein

Abstract:
Five different solvents were used to extract oil from silkwormpupa by Soxhlet′s extraction method. In order to increase the yield of protein extracted from silkwormpupa, the technology parameters were optimized by the method of dissolving with alkali and depositing with acid. The experiments of quadratic regression orthogonal rotary design were used to investigate the effects of some factors such as pH value, the ratio of material to solvent and extracting temperature on the yield of protein. The results were as follows: N-hexane was the best extracting solvent, the extraction percentage 28.9%; the optimum extracting conditions were that pH value 10, the ratio of material to solvent 1:12, extracting temperature 60℃ and extracting time 90 min. The yield of protein was 48.22% after double-extracted continuously. Moreover, the isoelectric point of pupa protein was about pH4.4,and the purity coefficient of the pupa protein was 95.2% after frozen-drying.
