超声结合原卟啉Ⅸ对艾氏腹水瘤细胞的杀伤作用

摘要:
通过台盼蓝拒染法,以细胞存活率为指标用扫描电镜观察超声激活原卟啉Ⅸ(Protoporphyrin Ⅸ, PP Ⅸ)对艾氏腹水肿瘤(Ehrlich ascites tumor, EAT)细胞的杀伤作用;采用硫代巴比妥酸法检测超声激活PP Ⅸ对EAT细胞膜脂质过氧化水平的影响,研究超声激活Ⅸ对EAT细胞的杀伤效应.结果显示,超声结合PP Ⅸ杀伤EAT细胞与超声强度、处理时间及PP Ⅸ药物浓度呈正相关,超声结合PP Ⅸ对EAT细胞的损伤显著高于单纯超声组,而单纯PP Ⅸ表现出无明显的细胞毒作用;超声激活PP Ⅸ后膜的脂质过氧化水平显著增加.实验表明超声激活PP Ⅸ对EAT细胞具有明显的协同杀伤效应,推测可能是超声激活PP Ⅸ增加了膜脂质过氧化量,从而使细胞受损,并且质膜可能是超声激活PP Ⅸ杀伤EAT细胞的主要靶点之一.

Abstract:
Typan blue exclusion method and the scanning electron microscope were used to analyze the viability of Ehrlich ascites tumor (EAT) cells in presence of protoporphyrin Ⅸ disodium salt (PP Ⅸ) ultrasonically activated.The level of lipid peroxidation was detected by using thiobarbituric acid reaction method.The cytotoxic effect of PP Ⅸ induced by ultrasound on isolated EAT cells were investigated.The results indicated that no significant effect could be checked out in the cells treated by PP Ⅸ.The cells treated by using ultrasound and PP Ⅸ were damaged more efficiently than the cells treated by using ultrasound alone.The damage could be enhanced with ultrasound intensity,treated time,and PP Ⅸ concentration.Comparing with the group untreated or treated by ultrasound alone,the level of lipid peroxides was notably increased in the group treated by ultrasound plus PP Ⅸ. In summary,PP Ⅸ could enhance the ultrasonically induced cell damage.The damage may be associated with the increased the level of lipid peroxidation.The cell membrane is probably one of the target organelles in this case.
