猪肝线粒体F1-ATP合酶的提取及检测方法

摘要:
采用超声和差速离心方法,研究了猪肝线粒体ATP合酶水溶性部分(F1-ATP合酶)的提取及检测方法.对初始提取液和纯化后的溶液分别进行酶比活力测量,并对常规聚丙烯酰胺电泳后的胶条进行胶内活性检测.结果表明,纯化后的F1-ATP合酶水解ATP比活力显著提高.

Abstract:
The extraction and detection of F1-ATP synthase from pig-liver mitochondria are investegated by using ultrasonic and differential centrifugal methods. The specific activity of F1-ATP synthase in both primary and purified solutions are measured first, then the in-gel enzymatic anylysis are made after polyacrylamide gel electrophoresis. The results show that the specific activity of F1-ATP synthase in purified solution is increased compared with that of the primary solution.
