运动员骨骼肌组织提取总RNA的新方法

摘要:
为了寻求一种从微量组织中高效又简便的提取骨骼肌总RNA方法,实验对常用RNA提取方法Trizol法进行了改进和简化,提取的总RNA质量进行了电泳和光密度值检测.实验结果显示,运动员微量骨骼肌组织提取的总 RNA 28S、18S和5S三条带清晰可见, RT-PCR 扩增的β-actin 参照基因条带具有特异性,说明该方法从微量组织可有效、简便地提取骨骼肌总 RNA,提取总 RNA 的得率和纯度均符合分子生物学进一步实验要求,为运动相关基因的分子生物学调控机制等研究提供了一定的方法参考.

Abstract:
To find an fast and effectual method for total RNA extracted from micro skeletal muscle tissues of athlete, the traditional Trizol method for total RNA extraction was improved, and the quality of the extracted total RNA was analysed by agarose electrophoresis and optical density test. The results show that total RNA extracted by the improved method has obvious three bands of 28S, 18S and 5S. The special bands expanded by RT-PCR is consistent with β-actin genes. RNA extracted by the method can meet the demands of gene cloning and of biomolecular research in yield and purity of total RNA. The improved method has an important reference to further study the regulating mechanisms about exercise-related genes expression.
