Abstract:
In order to select the optimum genotype for immature embryo culture, immature embryos from 12 common wheat genotypes including the improved cultivars and lines in Guanzhong district of Shaanxi province were cultured on both medium I and II, and the dedifferentiation characteristics was investigated. The results showed that time of callus formation and callus induction frequency were obviously different. The callus induction frequencies ranged from 65.9%~100.0%. The embryogenesis callus formation rates and the germination frequency had marked difference. Majority of materials had relative parallel in the tendency of higher or lower frequency through the actual frequency fluctuated between the two medias. The embryogenesis callus formation rates could be promoted by 6-BA. Additionally, the germination frequency could be increased too. In 12 tested genotypes, Xiaoyan 22, Shanmai 150 and Xiaoyan 54 displayed excellent tissue culture response due to the higher callus induction frequency and more excellent somatic embryogenetic competence.
