Abstract:
Pectinase was immobilized on anion-exchange resin by cross-linkage of glutaraldehyde. Effect of temperature, pH, time, amount of the enzyme, concentration of glutaraldehyde, crosslinking temperature and time were studied for the impact of immobilized pectinase. Then, the characteristics of immobilized enzyme was studied. The optimum conditions for the immobilization were as follows : 40℃, pH=5-5, 6 h, 0-75 mL pectinase/g resin.The concentration of glutaraldehyde was 0-1%, crosslinking temperature and time were 4℃ and 4h respectively. Under the optimal conditions, the activity recovery of immobilized pectinase can reach to more than 80%. The characteristics of immobilized enzyme were as follows:the optimum temperature and pH were 60℃ and 4-0 respectively. Moreover, the immobilized enzyme had a good operational stability.
