高效液相色谱化学发光法测定淫羊藿甙

摘要:
在碱性条件下,研究了淫羊藿甙与Co(Ⅱ)形成的1∶1配合物催化鲁米诺-H2O2体系产生化学发光.在此基础上建立了反相高效液相色谱(RP-HPLC) 分离柱后化学发光检测淫羊藿甙的新方法,并成功应用于安神补脑液中淫羊藿甙的测定.方法在0.5~20 μg/mL范围内与化学发光强度呈良好的线性关系,相关系数为0.9986,检出限为2×10-7g/mL(3σ).对2×10-6g/mL淫羊藿甙平行测定11 次,其相对标准偏差为4.5%.

Determination of icariin using HPLC with chemiluminescence detection

Abstract:
In alkaline solution, the complex of Cobalt (Ⅱ) with icariin can greatly catalyze the chemiluminescent reaction(CL) between Luminol and H2O2. Based on this, a novel HPLC-CL detection method for the determination of icariin was developed. The proposed method has been applied satisfactorily to the determination of icariin in Anshen Bunao mixture. The linear range for the determination of icariin was 0.5~20 μg/mL (r=0.998 6), and the detection limit was 2×10-7g/mL(3σ). The relative standard deviation was 4.5%(c=2×1010-6 g/mL, n=11).
