中国特有秦岭藤属药用植物的DNA条形码鉴定

摘要:
为筛选出能够准确鉴定秦岭藤属(Biondia Schltr.)药用植物的DNA条形码,选取5个叶绿体DNA片段,利用PWG-distance和Tree-building方法对中国中西部地区分布的秦岭藤属4个物种的14个个体进行物种分辨率评价。研究显示:大部分组合DNA片段的分辨率(25%~100%)显著高于单个DNA片段的分辨率(0~50%),尤其是matK+rbcL的核心DNA条形码组合,以及包含有这2个片段的3片段(matK+rbcL+trnH-psbA)核心DNA条形码组合,利用PWG-distance方法,其分辨率可达到100%。因此,可采用matK+rbcL或matK+rbcL+trnH-psbA的DNA条形码组合对秦岭藤属药用植物进行准确鉴定。

DNA barcoding identification of Chinese endemic medicinal plants in Biondia Schltr.

Abstract:
In order to find DNA barcodes that can accurately identify the medicinal plants of Biondia Schltr., the discrimination powers had been tested using five chloroplast DNA fragments in this study.14 individuals represented 4 Biondia species of the central and western regions of China were sampled. Two methods (PWG-distance and Tree-building) were used to conduct barcoding analysis.The results suggested that a single chloroplast DNA fragment always showed low rate of species discrimination(0~50%), however, most of the DNA fragment combinations had higher discrimination rates (25%~100%) than the single DNA fragment, especially for the combination of two core DNA barcodes (matK+rbcL) and three DNA fragments (matK+rbcL+trnH-psbA), whose discrimination rate could reach up to 100% when PWG-distance method was applied. Therefore, both combinations of matK+rbcL and matK+rbcL+trnH-psbA can be adopted for the identification of plants form Biondia Schltr..
