DNA barcoding identification of Chinese endemic medicinal plants in Biondia Schltr.

Abstract:
In order to find DNA barcodes that can accurately identify the medicinal plants of Biondia Schltr., the discrimination powers had been tested using five chloroplast DNA fragments in this study.14 individuals represented 4 Biondia species of the central and western regions of China were sampled. Two methods (PWG-distance and Tree-building) were used to conduct barcoding analysis.The results suggested that a single chloroplast DNA fragment always showed low rate of species discrimination(0~50%), however, most of the DNA fragment combinations had higher discrimination rates (25%~100%) than the single DNA fragment, especially for the combination of two core DNA barcodes (matK+rbcL) and three DNA fragments (matK+rbcL+trnH-psbA), whose discrimination rate could reach up to 100% when PWG-distance method was applied. Therefore, both combinations of matK+rbcL and matK+rbcL+trnH-psbA can be adopted for the identification of plants form Biondia Schltr..
