Prediction and identification for targetgenes of miR-424-5p associated with invasion and migrationof lung cancer

Abstract:
In order to further study the molecular mechanism of miR-424-5p involved in invasion and migration of lung cancer,screen for target genes of miR-424-5p associated with invasion and migration of lung cancer,the target genes of miR-424-5p associated with invasion and migration of lung cancer were investigated by literature review and analyzed using bioinformatics methods.The candidate target genes were screened out combined these two methods.The modified luciferase reporter vector pmirGLO reporters containing wild type and mutant 3′UTR sequences of the SOCS5 were constructed.These reporters and miR-424-5p mimics/NC were co-transfected into 293T cells respectively, and the relative luciferase activities were detected by dual luciferase reporter gene system.SIAH1 and SOCS5 were screened out as the candidate target genes of miR-424-5p by literatures review and bioinformatics analysis. Dual luciferase reporter gene system revealed that the relative luciferase activity of the reporter vector containing 3′UTR sequences of the SOCS5 and miR-424-5p mimic group was reduced to 46% of NC group.The relative luciferase activity of reporter vector containing of mutant 3′UTR sequences of the SOCS5 and miR-424-5p mimic group was not changed compared with that of reporter NC group. The results showed that SOCS5 was a potential target gene associated invasion and migrationof lung cancer of miR-424-5p.
