DNAzyme聚乙烯吡咯烷酮/Ru(bpy)<sup>2+</sup><sub>3</sub>/SiO<sub>2</sub>纳米粒子相互作用及铅(Ⅱ)的电化学发光分析方法研究

摘要:
合成了一种对单、双链DNA具有不同亲和能力的纳米粒子PVP/Silica/Ru(bpy)2+3。实验结果发现,Pb(Ⅱ)能够诱导DNAzyme结构的变化,纳米粒子PVP/Silica/Ru(bpy)2+3在Nafion/碳纳米管修饰电极表面具有电化学发光特性,据此建立了一种检测铅离子的电化学发光新方法。该方法具有免标记DNA探针、无需固定化DNAzyme、灵敏度高等特点。在最佳实验条件下,该方法对铅离子的检测限达0.5 nmol/L,实现了水样中Pb(Ⅱ)的电化学发光分析检测。

Abstract:
The PVP/Silica/Ru (bpy)2+3 nanoparticles were synthesized, which show different affinities for single and double-stranded DNA.It was found that changes of DNAzyme structure induced by Pb(Ⅱ) resulted in the change in electrochemiluminesence properties of nanoparticles in the Nafion/carbon nanotubes modified electrode. Based on this finding, a new electrochemiluminesence method for detecting Pb(Ⅱ) was developed.The method is a label-free DNA probe, no immobilized DNAzyme, and high sensitivity, etc. Under the optimal experimental conditions, this new electrochemiluminescence approach for Pb(Ⅱ) presented a detection limit of 0.5 nmol/L, and was successfully applied for detecting Pb(Ⅱ) in water samples.
