反相高效液相色谱测定太白泡沙参中β-谷甾醇的含量

摘要:
建立了反相高效液相色谱(RP-HPLC)测定太白泡沙参中β-谷甾醇含量的方法。在色谱柱为Kromasil C18 柱(5 μm,4.6 mm×250 mm)、紫外检测(波长210 nm)、色谱柱温度35 ℃,以甲醇为流动相且流速为0.7 mL/min的优化条件下,β-谷甾醇在 4×10-7~2×10-5g范围内线性关系良好,r=0.999 9,平均回收率为 99.70%,测定太白泡沙参中β-谷甾醇平均含量为0.134% 。此分析方法简便准确,可作为该产品的质量控制方法。

Abstract:
A method for the determination of β-sitosterol in A.potaninii Korsh from Shaanxi Taibai Mountainous area by reverse phase-high performance liquid chromatography(RP-HPLC) has been developed. The Kromasil C18 column (4.6 mm×250 mm,5 μm) was used for the separation with a mobile phase of pure methanol. The detected wavelength was 210 nm with a UV detector. The flow rate of mobile phase was 0.7 mL/min and the column temperature was 35 ℃ for the accurate determination of β-sitosterol. The method has a good linearity within a range of 4.0×10-7~2.0×10-5 g(r=0.999 9) under optimized conditions. The average recovery was 99.70%. The content of β-sitosterol in A.potaninii Korsh from Shaanxi Taibai Mountainous area was determined, and the average content of β-sitosterol was 0.134%. This method is simple and accurate to be used for the evaluating quality of A.potaninii Korsh.
