Abstract:
The extraction process for total saponins from kernel of Xanthoceras sorbifolia bunge seed were studied with ultrasound-assisted method. The separation and purification technology of total saponins from kernel of Xanthoceras sorbifolia bunge seed were discussed with macroporous resin, and its antioxidative activity of total saponins in vitro also was investigated. Firstly, using ethanol as extraction agent, the effect of the extract concentration and temperature, solid-liquid ratio, ultrasonic power on the total saponins extracting ratio were investigated by single factor and orthogonal experiments. The results show that: under the condition of the ethanol concentration of 70%, extraction temperature of 50 ℃, solid-liquid ratio of 1∶20, ultrasonic power of 140 W, the total content of saponins will get the highest ratio of 2.69%. Using XAD-16 macroporous resin to separated and purified the total saponins of Xanthoceras sorbifolia, the optimum conditions were as follows: the static adsorption and desorption time was 12 h and 4 h, eluent ethanol concentration is 70%, sample solution concentration is 0.12 mg/mL(pH=4), sample flow velocity is 10 mL/min, the proportion of sample volume and column volume is 1.5. The total saponins content of Xanthoceras sorbifolia was increased after saponin compounds was purified. Finally, using Vc as a control substance, the antioxidant activity of total saponins from kernel of Xanthoceras sorbifolia bunge seed was researched. The experimental results showed that the reducing power and hydroxy radical scavenging effect of total saponins was stronger than Vc, and the abilities of scavenging DPPH free radical and scavenging superoxide was less than Vc.
