产丝氨酸蛋白酶芽胞杆菌的鉴定及其生防作用

摘要:
为筛选丝氨酸蛋白酶产生菌用于生物防治,利用丝氨酸蛋白酶的特殊底物BApNA和特异性抑制剂PMSF确定产丝氨酸蛋白酶菌株,依据形态特征、生理生化特性和16S rDNA比对分析对菌株进行鉴定,用对峙法、抑菌圈法和液体振荡培养法观察抗植物病原真菌的作用,用96孔板法观察抗线虫作用。实验筛选到产丝氨酸蛋白酶的芽胞杆菌SNUB19,鉴定结果为枯草芽胞杆菌(Bacillus subtilis),该菌株发酵液的无菌滤液对尖孢镰刀菌等植物病原真菌有显著的抑制作用,对南方根结线虫有较强的致死效应。

Identification and biocontrol effect of a novel bacillus with producing serine protease

Abstract:
In order to select bacteria with producing serine protease for biological control, BApNA, the particular substrate of serine protease and selective inhibitor PMSF were used to confirm serine protease productivity. Morphological characteristics, physiological, biochemical characteristics and 16s rDNA comparison analysis were used for strain identification. Antagonistic method, inhibition zone method and liquid shake culture were introduced to observe the resistant effect of the bacteria on plant pathogenic fungi. 96-well plate assay was used to observe the resistant effect the bacteria on nematodes. SNUB19, a bacillus with producing serine protease was screened in the experiment and was identified as Bacillus subtilis. The fermentation broth of this strain is significantly resistant to plant pathogenic fungi and is relatively fatal to Meloidogyne incognita.
