红尾副鳅线粒体基因组序列测定与分析

摘要:
采用长PCR和嵌套PCR相结合的方法测定并注释了红尾副鳅Paracobitis variegatus(Dabry de Thiersant, 1874) 线粒体基因组全序列.结果表明:红尾副鳅线粒体基因组全长16 571 bp,A+T含量55.6%,基因构成及排列与其他硬骨鱼基本相同,包含37个基因和1个控制区(D-loop区).基因组结构紧密,除控制区外基因间仅存在63 bp的间隔,33 bp的重叠.13个蛋白质编码基因中,除了COI基因以GTG作为起始密码子,COII、COIII和Cyt b以不完整的T,ND4基因以不完整的TA作为终止密码子外,大都以典型的ATG作为起始密码子,以TAA或TAG作为终止密码子.22个tRNA基因的二级结构中,除了tRNASer(AGN)外,其余21个都可以折叠成典型的三叶草结构,tRNASer(AGN) 缺失了DHU臂,在相应位置上只形成一个环.预测了rRNA的二级结构,其中12S rRNA包含43个茎环结构,16S rRNA包含6个结构域,53个茎环结构.控制区包含1个终止相关序列TAS1、3个中央保守序列(CSB-F、CSB-D、CSB-E)、2个保守序列区(CSB-2、CSB-3).

Sequencing and analysis of the complete mitochondrial genome of Paracobitis variegatus

Abstract:
The complete mitochondrial genome (mitogenome) sequence of Paracobitis variegatus(Dabry de Thiersant, 1874) was sequenced using the strategy of sub-PCR after long-PCR amplification and annotated.The results showed that the entire mitochondrial genome of P.variegatus is 16 571 bp long with A+T content 55.6﹪.The mitochondrial genome organization of P.variegatus is the same with that in bony fish, which contains 37 genes and 1 control region (named D-loop).The genomic structure is arranged densely, besides D-loop control region, there is only 63 bp intergenic spacer, and 33 bp overlap between genes.Except for COI gene, which uses GTG as the initiation codon, all other protein-coding genes start with the typical ATG codon.The most of 13 protein-coding genes terminated with the typical TAA or TAG codons, while COⅡ, COIII and Cyt b were infered to have an incomplete termination codon T, and ND4 gene terminated with an incomplete termination codon TA.Most tRNA genes could form typical secondary structures except tRNASer(AGN), which had an absence of the DHU arm.The secondary structures of rRNA were predicted.There are 43 helices structures in 12S rRNA, and 6 domains, 53 helices structures in 16S rRNA.In addition, there are 1 terminal associated sequence named TAS1, 3 central conserved sequence blocks named CSB-F, CSB-E and CSB-D, 2 conserved sequence blocks named CSB-2, CSB-3 in the control region of P.variegatus.
