血卟啉单甲醚介导的声动力对U937细胞增殖影响

摘要:
研究了聚焦超声激活血卟啉单甲醚对人髓系白血病细胞株U937细胞的增殖抑制作用.通过MTT法检测了频率为1.1 MHz的聚焦超声激活血卟啉单甲醚作用于U937肿瘤细胞的存活率.MTT实验结果显示:声照辐射60 s、强度为2~7 W/cm2的超声对U937细胞的抑制作用呈强度依赖性;单纯血卟啉单甲醚组与对照组相比无显著性差异(P>0.05);同等条件下,2 W/cm2超声结合10 ng/L血卟啉单甲醚组对U937肿瘤细胞有明显的协同杀伤作用,与对照组相比有极显著性差异(P<0.01).这些结果表明,超声联合血卟啉单甲醚声动力能够特异性抑制U937细胞生长,血卟啉单甲醚有望成为一种新型声敏剂应用于白血病的声动力治疗.

The sonodynamic effect of HMME on the proliferation of U937 cells

Abstract:
The present study aims to investigate the cytotoxic effect of focused ultrasound combining HMME on human leukemia U937 cells. Tumor cells suspended in serum-free RPM1640 were exposed to ultrasound with 1 W/cm2 and 1.1 MHz for 60 seconds in the presence and absence of HMME. The viability of cells was determined by MTT. MTT results showed that the treatment with ultrasound (1.1 MHz, 2~7 W /cm2, 60 s) alone significantly inhibited the cell proliferation of the U937 cells in a intensity dependence,while the HMME (10~100 ng/L) alone showed no statistically significant differences with the control groups (P>0.05); Under the same conditions, the synergistic effect of 2 W /cm2 ultrasound combined with HMME showed significant cell killing compared with control (P
