Evaluation on the invitro anti-tumor activity of three kinds of Coprinus comatus polysaccharides by MTT-assay

Abstract:
The in vitro anti-tumor activity of three kinds of Coprinus comatus polysaccharides were tested by MTT assay. The Coprinus comatus polysaccharides were isolated by ultrasonic(UCP-3), then carboxymethylated Coprinus comatus polysaccharide(CWCP) and sulfated Coprinus comatus polysaccharide(SWCP) were obtained respectively. The analyses of anti-tumor activity of polysaccharides UCP-3, SWCP and CWCP on human leukemia K562 cells in vitro were carried out. The effect of in vitro anti-tumor activity of Coprinus comatus polysaccharides extracted through different processes was investigated. The results show that three kinds of Coprinus comatus polysaccharides all have inhibitory action on K562 cells. The ultrasound can induce the connection pattern change of monosaccharides thus resulting the change of bioactivity. The inhibition effect of SWCP and CWCP are obvious enhanced, so sulfation and carboxymethylation modifications can improve the biological activity.
